Overview

PVP003 is a tablet formulation of **TAK-062** (formerly known as Kuma062), a recombinant, computationally engineered gluten-degrading enzyme (glutenase). Developed by PvP Biologics and later acquired by Takeda, the drug is designed to treat celiac disease by breaking down immune-reactive gluten peptides in the stomach before they reach the small intestine. The enzyme is a highly potent prolyl endopeptidase (PEP) derived from *Alicyclobacillus sendaiensis* (kumamolisin-As), engineered to be active at the low pH of the stomach and highly resistant to proteolysis by pepsin. Clinical studies, including NCT03701555, have evaluated PVP003 alongside other formulations (PVP001 liquid and PVP002 capsules) to determine safety, tolerability, and efficacy in degrading gluten in healthy volunteers and patients with celiac disease.
