TAK‑071 is an investigational small molecule that acts as a positive allosteric modulator of the muscarinic acetylcholine receptor M1 (M1R). It was developed by Takeda for the treatment of cognitive impairment associated with Parkinson's disease and Alzheimer's disease. TAK‑071 selectively enhances M1 receptor activity in the presence of acetylcholine, aiming to improve cognitive function without causing significant cholinergic side effects. Clinical studies have shown that while TAK‑071 did not significantly improve gait variability in Parkinson's disease patients with cognitive impairment, it did demonstrate improvement in composite cognitive scores compared to placebo. The drug has been well-tolerated in clinical trials but development for both Parkinson’s and Alzheimer’s diseases has been discontinued[2][3][8][10].
