TAK-137 is a novel small molecule AMPA receptor potentiator developed by Takeda. It acts as a positive allosteric modulator of the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor, an ionotropic glutamate receptor. Unlike direct agonists, TAK-137 has little intrinsic agonistic activity but enhances AMPA receptor function, leading to increased glutamatergic neurotransmission. Preclinical studies demonstrated that TAK-137 produces antidepressant-like effects and procognitive benefits without causing psychotomimetic or seizure-inducing side effects. It was investigated for the treatment of psychiatric and neurological disorders such as major depressive disorder (including treatment-resistant depression), schizophrenia (targeting cognitive and negative symptoms), and attention-deficit hyperactivity disorder. Despite promising preclinical results showing improved cognition and social functioning in animal models, clinical development was discontinued after phase 1 trials[1][2][4][6].
