Overview

TAK-186 is a conditionally active bispecific T-cell engager (TCE) designed to target both the epidermal growth factor receptor (EGFR) and CD3 on cytotoxic T lymphocytes. It is engineered using COBRA (Conditional Bispecific Redirected Activation) technology to remain inactive in healthy tissues and become activated within the tumor microenvironment through protease-mediated cleavage. Upon activation, TAK-186 dimerizes and binds simultaneously to EGFR on tumor cells and CD3 on T-cells, redirecting T-cell cytotoxicity specifically toward EGFR-expressing cancer cells. This mechanism aims to enhance anti-tumor efficacy while minimizing off-tumor toxicity. The drug is being developed primarily for advanced or metastatic solid tumors that are unresectable or inoperable[4][6][7][8][10].
