TAK-214 (formerly HIL-214) is a bivalent virus-like particle (VLP) vaccine candidate designed to prevent acute gastroenteritis caused by norovirus infection. The vaccine contains recombinant VLP antigens representing two major norovirus genotypes: the GI.1 (Norwalk virus) strain and a consensus GII.4 sequence (GII.4c) derived from multiple variants. These VLPs mimic the external structure of the norovirus capsid but lack the viral genome, rendering them non-infectious while remaining highly immunogenic. The formulation is typically adjuvanted with aluminum hydroxide to enhance the immune response. Originally developed by Takeda, the program was licensed to HilleVax for further clinical development, targeting both pediatric and adult populations to address the significant global burden of norovirus-related illness.
