**TAK-279 + midazolam** is a combination of TAK-279 (zasocitinib), a highly selective oral tyrosine kinase 2 (TYK2) inhibitor, and midazolam, a short-acting benzodiazepine used primarily as a sedative, anxiolytic, and for procedural sedation. TAK-279 acts by selectively inhibiting TYK2, a kinase involved in the signaling of key cytokines implicated in inflammatory and autoimmune diseases, with next-generation selectivity that avoids toxicities associated with broader JAK inhibition. Midazolam acts as a positive allosteric modulator of the gamma-aminobutyric acid type A (GABA-A) receptor, enhancing inhibitory neurotransmission in the central nervous system. The combination has been used in clinical pharmacokinetic interaction studies to evaluate the effect of TAK-279 on the metabolism of midazolam, a probe substrate for cytochrome P450 (CYP) 3A4, in healthy adults. These studies assess whether TAK-279 alters the pharmacokinetics or tolerability of midazolam when co-administered[1][3][5].
