Overview

TAK-280 is an investigational bi-specific monoclonal antibody developed using the COBRA (Conditional Bispecific Redirected Activation) platform. It is designed as a conditionally active T-cell engager that targets B7 homolog 3 protein (B7-H3, also known as CD276), which is highly expressed in a variety of solid tumors including metastatic castration-resistant prostate cancer (mCRPC), non-small cell lung cancer, head and neck squamous cell carcinoma, bladder cancer, small-cell lung cancer, cutaneous melanoma, and metastatic uveal melanoma. In its prodrug form, TAK-280 binds to B7-H3 but not to CD3ε. Upon exposure to the protease-rich tumor microenvironment (TME), it undergoes protease-mediated activation resulting in formation of active dimers capable of binding both B7-H3 on tumor cells and CD3 on T-cells. This conditional activation leads to localized T-cell engagement and cytotoxic antitumor response while limiting toxicity to normal tissues. The drug is administered intravenously and remains under clinical investigation for unresectable locally advanced or metastatic cancers.
