TAK-441

TAK‑441 is an orally bioavailable small molecule inhibitor of the Hedgehog (Hh) signaling pathway. It acts as a potent and selective antagonist of Smoothened (Smo), a key cell surface co-receptor in the Hh pathway. By binding to and inhibiting Smo, TAK‑441 suppresses downstream signaling events including Gli1 mRNA expression and tumor growth in preclinical models. The drug has demonstrated antitumor activity in various solid tumor models by disrupting paracrine Hh signaling between tumor cells and stroma. Originally developed by Millennium (a Takeda company), it has been investigated for cancers such as prostate cancer and pancreatic cancer[1][3][4][5][7][8].
