TAK-505 is an investigational immune cell-directed antibody-drug conjugate (iADC) developed by Takeda for the treatment of advanced solid tumors. It consists of a human IgG1 monoclonal antibody targeting C-C chemokine receptor 2 (CCR2) conjugated to a STING (Stimulator of Interferon Genes) agonist payload via a protease-cleavable maleimide linker. The drug is designed to selectively deliver the STING agonist to CCR2-expressing myeloid cells, such as tumor-associated macrophages and myeloid-derived suppressor cells, within the tumor microenvironment. By activating the STING pathway locally, TAK-505 aims to enhance type I interferon production and stimulate both innate and adaptive immune responses against cancer cells. Clinical development in a Phase 1/2 trial (NCT05070247) for various solid tumors, including gastric adenocarcinoma, colorectal cancer, and non-small cell lung cancer, was reportedly terminated due to dose-limiting toxicities.
