TAK-580 + plozalizumab + vedolizumab is a combination therapy of three agents: - **TAK-580 (tovorafenib/MLN2480)**, a pan-RAF inhibitor targeting the RAS-RAF-MEK-ERK pathway that disrupts RAF dimerization and inhibits tumor cell proliferation, with activity shown against various solid and hematological tumors including glioma and melanoma. - **Plozalizumab**, a monoclonal antibody developed to target CCR2 (CC chemokine receptor 2), primarily investigated for inflammatory or fibrotic indications; information on its oncology application is limited. - **Vedolizumab**, a recombinant humanized IgG1 monoclonal antibody against the α4β7 integrin; it blocks lymphocyte trafficking into the gastrointestinal tract and exerts gut-selective anti-inflammatory effects, clinically approved for ulcerative colitis and Crohn’s disease.
