Overview

SHP639 (also known as TAK-639) is a small molecule agonist of the natriuretic peptide receptor B (NPR-B) that was developed by Takeda, following its acquisition of Shire, for the treatment of primary open-angle glaucoma and ocular hypertension. As a C-type natriuretic peptide (CNP) analog, it targets NPR-B receptors located in the trabecular meshwork and ciliary body of the eye. Activation of these receptors stimulates the production of cyclic guanosine monophosphate (cGMP), which facilitates the outflow of aqueous humor and reduces intraocular pressure. SHP639 was evaluated in Phase 1 clinical trials as a topical ophthalmic solution but was subsequently discontinued from Takeda's development pipeline in 2021.
