Overview

**TAK-659 + ibrutinib** is an investigational drug combination developed for the treatment of B-cell malignancies, particularly chronic lymphocytic leukemia (CLL) and non-Hodgkin lymphoma. TAK-659 (mivavotinib) is a small molecule inhibitor targeting spleen tyrosine kinase (SYK) and FLT3, disrupting B-cell receptor (BCR) signaling and cellular proliferation. Ibrutinib is an orally active small molecule Bruton's tyrosine kinase (BTK) inhibitor that blocks BCR signaling downstream, impairing tumor growth and survival. Preclinical studies show the combination induces synergistic cytotoxicity and apoptosis in malignant B cells, potentially overcoming resistance mechanisms seen with ibrutinib monotherapy.
