TAK-676 + carboplatin + 5-fluorouracil is a multi-agent investigational combination consisting of TAK-676 (also known as dazostinag), carboplatin, and 5-fluorouracil, under clinical evaluation for the treatment of advanced or metastatic solid tumors. **TAK-676** is a synthetic small molecule agonist of the Stimulator of Interferon Genes (STING) pathway, designed for systemic intravenous administration. It activates STING signaling, inducing type I interferon production, enhancing both innate and adaptive immunity, and promoting antitumor immune responses[1][6]. **Carboplatin** is a platinum-based chemotherapeutic that damages DNA through the formation of intra- and interstrand DNA crosslinks, leading to apoptosis in rapidly dividing cells. **5-Fluorouracil** is a pyrimidine analog antimetabolite that inhibits thymidylate synthase and incorporates into RNA and DNA, disrupting cancer cell replication. This combination is intended to harness both direct cytotoxic effects and immune activation to overcome resistance mechanisms and improve efficacy in cancers such as head and neck squamous cell carcinoma and other advanced solid tumors.
