TAK-683 is a synthetic nonapeptide analog of the C-terminal decapeptide of kisspeptin (metastin), developed as a potent and metabolically stable agonist of the KISS1 receptor (also known as GPR54 or metastin receptor). TAK-683 acts as a full agonist at KISS1R and has been investigated for its potential to suppress reproductive hormone secretion, including testosterone, luteinizing hormone (LH), and follicle-stimulating hormone (FSH), primarily for the treatment of hormone-dependent diseases such as prostate cancer. Preclinical and clinical studies have demonstrated that TAK-683 initially stimulates LH, FSH, and testosterone release, followed by profound suppression of these hormones and testosterone to castrate levels with continuous administration, suggesting utility in androgen deprivation therapy. It has also been studied for its effects on reproductive physiology in animal models.
