Overview

TAK-715 + methotrexate is a combination of two small molecule drugs. TAK-715 is an orally bioavailable, potent and selective inhibitor of p38 mitogen-activated protein kinase alpha (p38 MAPKα), with an IC50 of 7.1 nM[1][2][3][4]. It also inhibits Wnt/β-catenin signaling and several other kinases, including CK1δ/ε[1][5]. TAK-715 was developed as an anti-inflammatory and anti-rheumatoid arthritis agent by Takeda, targeting the pro-inflammatory cytokine signal pathway to reduce TNF-alpha production[4][6]. Methotrexate is a well-established antimetabolite and antifolate drug widely used in the treatment of rheumatoid arthritis and various cancers. The combination was investigated for enhanced efficacy in rheumatoid arthritis but development of TAK-715 was discontinued after Phase II trials due to insufficient efficacy for further development[4].
