TAK-733 + alisertib is an investigational combination therapy consisting of two small molecule inhibitors developed for the treatment of advanced nonhematologic malignancies and other solid tumors. TAK-733 is a selective inhibitor of MEK (mitogen-activated protein kinase kinase), targeting the MAPK/ERK pathway involved in cell proliferation and survival. Alisertib (also known as MLN8237) is a selective Aurora A kinase inhibitor that disrupts mitotic progression and induces cell cycle arrest. The rationale for combining these agents lies in their complementary mechanisms—MEK inhibition suppresses oncogenic signaling, while Aurora A kinase inhibition impairs mitosis—potentially resulting in synergistic antitumor effects such as enhanced apoptosis and tumor growth inhibition. This combination has been evaluated in phase 1 clinical trials for patients with advanced solid tumors, including triple-negative breast cancer and other nonhematologic malignancies[1][3][4][5][6].
