Overview

TAK-783 is a small molecule inhibitor of spleen tyrosine kinase (Syk) that was developed by Takeda for the treatment of rheumatoid arthritis. Syk is a non-receptor cytoplasmic tyrosine kinase that plays a critical role in the signaling pathways of various inflammatory cells, including B cells and mast cells, by mediating downstream signaling from immunoreceptors such as the B-cell receptor (BCR) and Fc receptors. By inhibiting Syk, TAK-783 was designed to reduce the production of pro-inflammatory cytokines and the activation of immune cells involved in joint destruction. The drug was evaluated in Phase 2 clinical trials (e.g., NCT01066091) as an oral, once-daily treatment in combination with methotrexate for patients with active rheumatoid arthritis. However, Takeda discontinued the development of TAK-783 around 2012. While some early pipeline documents misidentified the target as GPR15, later official reports and scientific abstracts confirmed its mechanism as Syk inhibition.
