Overview

TAK-948 is an investigational, allogeneic (off-the-shelf) T-cell therapy derived from induced pluripotent stem cells (iPSCs) and engineered with a chimeric antigen receptor (CAR) targeting Glypican-3 (GPC3). Developed by Takeda in collaboration with the Center for iPS Cell Research and Application (CiRA) at Kyoto University, TAK-948 was designed to treat GPC3-positive solid tumors, most notably hepatocellular carcinoma (HCC). By utilizing iPSC technology, the therapy provides a standardized, renewable source of T cells, potentially overcoming the manufacturing hurdles and patient-to-patient variability associated with autologous CAR-T therapies. The therapy involves the differentiation of iPSCs into T cells that are then modified to express a CAR specific for GPC3, a cell-surface oncofetal antigen highly expressed in liver cancer. In 2023, Takeda terminated the Phase 1 clinical trial (NCT04683003) and discontinued the development of TAK-948 as part of a strategic prioritization of its oncology pipeline.
