Overview

**TAK-981 + pembrolizumab** is an investigational combination therapy evaluated for select advanced or metastatic solid tumors. TAK-981 is a first-in-class small molecule inhibitor targeting the SUMOylation pathway via inhibition of the SUMO-activating enzyme, resulting in a SUMO-TAK-981 adduct that blocks enzyme function. This pharmacologically reactivates type I interferon signaling, promotes T cell activation, enhances macrophage phagocytosis and NK cell cytotoxicity, disrupts Treg cell immune suppression, and inhibits tumor cell proliferation and angiogenesis. Pembrolizumab is a well-established anti-PD-1 monoclonal antibody checkpoint inhibitor that enhances anti-tumor immune responses by preventing PD-1-mediated T cell inhibition. The combination aims to maximize immune activation against tumor cells by simultaneous blockade of immune checkpoints (pembrolizumab) and stimulation of innate and adaptive immune pathways (TAK-981)[1][5].
