Talacotuzumab is a humanized IgG1 monoclonal antibody that targets the alpha subunit of the interleukin-3 receptor (CD123, also known as IL3RA). It has been engineered to have increased affinity for CD16 on natural killer cells, thereby enhancing antibody-dependent cellular cytotoxicity (ADCC) against CD123-expressing cells. Talacotuzumab was developed primarily for hematologic malignancies such as acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS), with clinical trials investigating its use in patients not eligible for intensive chemotherapy. The drug acts by binding to CD123, inhibiting downstream IL-3 signaling and promoting immune-mediated destruction of malignant cells[1][2][4][5].
