talampanel + temozolomide

Talampanel + temozolomide is a combination therapy investigated for the treatment of newly diagnosed glioblastoma multiforme. Talampanel is an oral, noncompetitive antagonist of the alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor, a subtype of glutamate receptor implicated in tumor proliferation and migration. Temozolomide is an oral alkylating agent that induces DNA damage and cell death, widely used as standard chemotherapy for glioblastoma. The combination has been studied alongside radiation therapy to assess whether AMPA receptor blockade by talampanel can enhance the efficacy of standard chemoradiation with temozolomide.
