Overview

Talaporfin sodium is a second-generation, water-soluble chlorin-based photosensitizer used in photodynamic therapy (PDT). Developed by Light Sciences Oncology as part of the Litx platform, the drug is administered intravenously and subsequently activated by low-intensity light from percutaneously implanted interstitial light-emitting diodes (LEDs). This activation triggers the production of singlet oxygen and other reactive oxygen species, which induce localized tumor cell apoptosis and microvascular shutdown. The primary clinical focus for Light Sciences Oncology was the treatment of unresectable hepatocellular carcinoma (HCC), where it was evaluated in a Phase 3 trial (NCT00389389) against standard-of-care therapies like TACE and RFA. However, development by the company was halted following financial difficulties and the termination of its pivotal trials. The drug remains approved in Japan for the treatment of certain cancers, such as glioma and esophageal cancer, under the brand name Laserphyrin.
