Overview

Talaporfin sodium + FOLFOX4 is a combination therapeutic regimen investigated for the treatment of colorectal cancer liver metastases (CRLM). This approach integrates photodynamic therapy (PDT) with systemic chemotherapy. Talaporfin sodium is a second-generation, water-soluble photosensitizer derived from chlorophyll that, when activated by a 664 nm diode laser, generates reactive oxygen species (primarily singlet oxygen) to induce localized tumor necrosis. FOLFOX4 is a standard chemotherapy regimen consisting of oxaliplatin, leucovorin (folinic acid), and 5-fluorouracil, which provides systemic anti-tumor activity by inhibiting DNA synthesis and repair. The combination is typically employed by administering talaporfin sodium intravenously followed by laser irradiation of liver lesions during surgery or via interstitial fibers, while the patient concurrently receives systemic FOLFOX4 to address micrometastatic disease and reduce local recurrence.
