Overview

Talazoparib + avelumab is an investigational combination therapy for cancer, particularly in patients with tumors harboring BRCA1, BRCA2, or ATM mutations. Talazoparib is an oral small molecule inhibitor of poly ADP-ribose polymerase (PARP), which impairs DNA repair in tumor cells and leads to cell death. Avelumab is an intravenous monoclonal antibody targeting programmed death ligand 1 (PD-L1), blocking its interaction with PD-1 and B7.1 receptors on T cells to restore anti-tumor immune responses. The combination aims to exploit synthetic lethality from PARP inhibition alongside immune checkpoint blockade to enhance anti-cancer efficacy[1][3][4][5][6].
