TALEN PD-1 knockout refers to a therapeutic approach where Transcription Activator-Like Effector Nucleases (TALENs) are used to permanently disrupt the PDCD1 gene, which encodes the Programmed Cell Death Protein 1 (PD-1) receptor. This modification is typically performed ex vivo on autologous or allogeneic T cells. By knocking out PD-1, the T cells are rendered resistant to PD-L1-mediated immunosuppression within the tumor microenvironment, thereby enhancing their anti-tumor activity and persistence. This strategy is primarily investigated in the context of adoptive cell therapies for various malignancies, including esophageal cancer and other solid tumors.
