taletrectinib

Taletrectinib is an oral small molecule tyrosine kinase inhibitor (TKI) that selectively targets ROS1 and tropomyosin receptor kinases (TRK), including TRKA, TRKB, and TRKC. It is designed to inhibit both wild-type ROS1 and resistance mutations such as G2032R. Taletrectinib blocks the activity of these kinases in cancer cells with ROS1 rearrangements or fusions, thereby inhibiting downstream signaling pathways that drive uncontrolled cell proliferation and tumor growth. The drug has demonstrated high systemic and intracranial response rates in patients with locally advanced or metastatic ROS1-positive non-small cell lung cancer (NSCLC), including those who have developed resistance to prior TKI therapies or present with central nervous system metastases. Developed by Nuvation Bio, taletrectinib was approved by the FDA in June 2025 for this indication under the brand name Ibtrozi in the US and Dovbleron in China[2][6][7][8].
