Talineuren (formerly known as Neurotone) is a liposomal formulation of monosialotetrahexosylganglioside (GM1), a naturally occurring glycosphingolipid that is highly concentrated in the mammalian central nervous system. Developed by Taliun AG, Talineuren is designed to enhance the delivery of GM1 across the blood-brain barrier to provide neuroprotective and neurorestorative effects in neurodegenerative disorders, particularly Parkinson's disease. GM1 ganglioside is known to modulate several critical neuronal processes, including neurotrophic factor signaling (acting as a co-receptor or modulator for TrkA and other receptors), ion pump activity (such as Na+/K+-ATPase), and calcium homeostasis. The Phase 1 NEON trial (NCT04976127) demonstrated that weekly intravenous infusions of Talineuren were generally safe and well-tolerated in patients with Parkinson's disease, with exploratory results suggesting improvements in both motor and non-motor symptoms.
