Talviraline is a second-generation quinoxaline-based small molecule that acts as a non-nucleoside reverse transcriptase inhibitor (NNRTI) targeting HIV-1. It was developed for the treatment of HIV infections and demonstrated potent inhibition of HIV-1-induced cell killing and viral replication in human cell lines and primary cells. Talviraline reached phase II clinical trials where it showed pronounced acute suppression of HIV replication both as monotherapy and in combination with zidovudine (AZT). However, its development was discontinued before approval[1][2][3][4][5][7][8].
