TAM558 (also known as Cytolysin-TAM558) is a synthetic tetrapeptidic analogue of natural tubulysins developed by Oncomatryx. It serves as a potent cytotoxic payload for antibody-drug conjugates (ADCs), including OMTX705, OMTX703, and OMTX707. TAM558 functions by disrupting microtubule dynamics, which inhibits cell division and triggers apoptosis in cancer cells. A key advantage of TAM558 is that it is not a substrate for Multiple Drug Resistance (MDR) proteins, potentially overcoming resistance mechanisms that affect other microtubule-targeting agents. It also demonstrates anti-angiogenic activity. The molecule was developed in collaboration with Tube Pharmaceuticals, which was acquired by Oncomatryx in 2023.
