A combination therapy consisting of tamoxifen, a selective estrogen receptor modulator (SERM), and tipifarnib, a farnesyltransferase inhibitor (FTI). This combination has been studied for the treatment of tamoxifen-resistant metastatic breast cancer. Preclinical data showed synergistic antiproliferative effects, with the combination producing greater G1 cell cycle arrest than either drug alone, marked inhibition of cyclin D1, and induction of the cell cycle inhibitor p27(kip1).
