Overview

Tamrintamab pamozirine is an investigational antibody-drug conjugate (ADC) that targets DPEP3 (dipeptidase 3), a glycosylphosphatidylinositol-anchored membrane protein from the DPEP family. The ADC consists of a proprietary monoclonal antibody directed against DPEP3, conjugated to a cytotoxic payload based on pyrrolobenzodiazepine. Upon binding to DPEP3-expressing tumor cells, the ADC is internalized and releases its cytotoxic agent, leading to cell death—likely through inhibition of DNA replication, although the precise mechanism remains under investigation. Tamrintamab pamozirine was primarily developed for platinum-resistant ovarian carcinoma and has been evaluated in early-phase clinical trials; however, development has been discontinued following limited efficacy results[1][2][3][4][5][6][7].
