Tandem CD33-CLL1 CAR-T is an investigational chimeric antigen receptor (CAR) T-cell therapy engineered to simultaneously target two myeloid-associated antigens: CD33 (Myeloid cell surface antigen CD33) and CLL1 (C-type lectin domain family 12 member A, also known as CLEC12A). This dual-targeting approach is designed to overcome antigen escape and improve the eradication of acute myeloid leukemia (AML) blasts and leukemic stem cells. The therapy was developed through a collaboration involving the University of Science and Technology of China, The Second Affiliated Hospital of Anhui Medical University, Soochow University, and Legend Biotech. While preclinical models showed potent anti-leukemic activity and minimal hematopoietic toxicity, an investigator-initiated clinical trial in China led to its removal from Legend Biotech's development pipeline due to insufficient CAR-T cell expansion and clinical efficacy.
