Overview

This is a combination therapy consisting of three agents: tandutinib, bevacizumab, and temozolomide. Tandutinib (MLN518) is an oral small molecule inhibitor targeting receptor tyrosine kinases including Fms-like tyrosine kinase 3 (FLT3), Platelet-derived growth factor receptor alpha (PDGFRα/β), and Mast/stem cell growth factor receptor Kit (c-Kit). Bevacizumab is a monoclonal antibody that inhibits Vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis. Temozolomide is an oral alkylating agent that induces DNA damage leading to tumor cell death and also impairs angiogenic processes in glioma models. This combination has been investigated primarily for the treatment of glioblastoma multiforme (GBM), aiming to target both tumor cells directly and the supporting vasculature through complementary mechanisms[1][2][4].
