Tanezumab is a humanized monoclonal antibody of the IgG2 class that targets and inhibits nerve growth factor (NGF), a neurotrophin involved in the regulation of pain signaling. By binding to NGF, tanezumab prevents its interaction with high-affinity (TrkA) and low-affinity (p75) receptors on nociceptive neurons, thereby interrupting pain transmission from peripheral tissues to the central nervous system. This mechanism is distinct from traditional analgesics such as opioids or NSAIDs, offering a novel approach for managing chronic pain conditions. Tanezumab was originally discovered by Rinat Neuroscience and later acquired and developed by Pfizer; Eli Lilly also became a co-development partner. The drug has been primarily investigated for moderate-to-severe osteoarthritis pain and chronic low back pain in patients who have not responded adequately to other treatments[1][4][5][6][7].
