Tanomastat is a synthetic small molecule and biphenyl matrix metalloproteinase inhibitor (MMPI), developed primarily as an antineoplastic agent. It selectively inhibits matrix metalloproteinases MMP‑2, MMP‑3, and MMP‑9, enzymes involved in extracellular matrix degradation. By inhibiting these targets, tanomastat was designed to block tumor angiogenesis, growth, invasion, and metastasis. It has also shown antiangiogenic and antimetastatic properties in vivo. Tanomastat was investigated for the treatment of several cancers—including ovarian cancer (as maintenance therapy after chemotherapy), pancreatic cancer, lung cancer—as well as osteoarthritis. Despite reaching phase III clinical trials for some indications, all development was discontinued following negative results in pivotal studies[1][2][6][7].
