Overview

Tantalum nanoparticles (Ta-NPs) are an investigational high-atomic-number (high-Z) metallic nanomaterial designed to function as a radiosensitizer in oncology. Tantalum (Z=73) possesses a high X-ray attenuation coefficient, allowing the nanoparticles to absorb ionizing radiation more effectively than biological tissue. Upon irradiation, Ta-NPs undergo photoelectric and Compton effects, leading to the emission of secondary ionizing species such as Auger electrons and photoelectrons. These secondary emissions increase the local radiation dose and promote the generation of reactive oxygen species (ROS), thereby enhancing DNA damage and cell death within the tumor microenvironment. This approach aims to improve the efficacy of radiotherapy while sparing adjacent healthy organs, which is particularly critical in the management of retroperitoneal sarcomas. The agent is currently being evaluated in Phase 1 clinical trials by West China Hospital for the treatment of locally recurrent retroperitoneal dedifferentiated liposarcoma and leiomyosarcoma.
