TAP311 is a small molecule inhibitor of cholesteryl ester transfer protein (CETP), developed by Novartis. It is an achiral, carboxylic acid-bearing piperidine-based compound with a core structure distinct from other CETP inhibitors. TAP311 potently inhibits CETP activity in human plasma (IC50 = 62 nM), aiming to modulate lipid metabolism and improve dyslipidemia profiles. Preclinical studies demonstrated robust efficacy in animal models and favorable pharmacokinetics, with reduced lipophilicity compared to anacetrapib and no increase in aldosterone secretion as seen with torcetrapib. Clinical development reached Phase 1 trials for dyslipidemia but did not advance further[1][2][4].
