Overview

Tapinarof is a naturally derived, nonsteroidal small molecule that acts as an aryl hydrocarbon receptor (AhR) agonist. It is used topically for the treatment of plaque psoriasis in adults and atopic dermatitis in adults and children aged 2 years and older. Tapinarof modulates immune responses by activating AhR, which leads to downregulation of pro-inflammatory cytokines (notably Th2), upregulation of skin barrier proteins, reduction of oxidative stress via activation of NRF2 pathways, and normalization of skin homeostasis. The drug has demonstrated significant efficacy in clinical trials for both plaque psoriasis and atopic dermatitis with a favorable safety profile[1][3][5][6][8].
