Overview

Taplucainium chloride (NOC-100) is a small molecule, permanently charged sodium channel blocker developed by Nocion Therapeutics. It is designed to selectively target and silence activated nociceptors (sensory neurons) involved in cough and pain without affecting other neurons. Unlike traditional local anesthetics, taplucainium chloride is a quaternary ammonium compound that cannot cross cell membranes on its own; instead, it enters neurons through large-pore ion channels (such as TRPV1 or P2X receptors) that are opened during inflammation or irritation. Once inside, it blocks voltage-gated sodium channels from the intracellular side, providing targeted antitussive effects. It is primarily being investigated for the treatment of refractory or unexplained chronic cough.
