Overview

Tariquidar is a third-generation, potent, and specific inhibitor of P-glycoprotein (Pgp), an ATP-dependent efflux transporter that mediates multidrug resistance in cancer cells. By inhibiting Pgp, tariquidar can increase the intracellular concentration of chemotherapeutic agents such as taxanes and platinum compounds. Paclitaxel is a microtubule-stabilizing agent that disrupts cell division by promoting tubulin polymerization, leading to apoptosis in rapidly dividing cells. Carboplatin is a platinum-based chemotherapy drug that forms DNA adducts and crosslinks, resulting in inhibition of DNA synthesis and cell death. The combination of these three drugs aims to overcome drug resistance mechanisms mediated by Pgp while maximizing cytotoxic effects against cancer cells. This regimen has been investigated primarily for advanced or refractory solid tumors including non-small cell lung cancer and ovarian cancer[1][2][4][5][6].
