Overview

**Tarlatamab + durvalumab** is an investigational combination therapy studied in extensive-stage small cell lung cancer (ES-SCLC), particularly following or as maintenance after platinum-based chemotherapy. **Tarlatamab** is a bispecific T-cell engager (BiTE®) monoclonal antibody that targets DLL3 (delta-like ligand 3) on tumor cells and CD3 on T cells, facilitating a redirected T-cell–mediated immune attack against DLL3-expressing cancer cells. **Durvalumab** is a monoclonal antibody immune checkpoint inhibitor targeting programmed death ligand 1 (PD-L1), which blocks its immunosuppressive signal and enhances anti-tumor immune response. The combination aims to leverage both direct T-cell engagement (by tarlatamab) and checkpoint inhibition (by durvalumab) to improve outcomes in ES-SCLC. The combination is under evaluation in phase 3 trials for ES-SCLC and is not approved for any other indication. Tarlatamab was developed by Amgen and durvalumab by AstraZeneca[1][2][7][9].
