TAS-119 + paclitaxel is a combination therapy of TAS-119, a selective orally active small molecule inhibitor of Aurora kinase A, and paclitaxel, a widely used microtubule-stabilizing chemotherapeutic agent. TAS-119 inhibits Aurora A kinase, a key regulator of mitosis; overexpression of Aurora A can confer resistance to taxanes such as paclitaxel. Preclinical and early-phase clinical studies indicate that TAS-119 enhances the antitumor efficacy of paclitaxel in various cancer models, including paclitaxel-resistant tumors, without significantly increasing taxane-related toxicities. The combination has undergone clinical investigation for advanced solid tumors, particularly breast and ovarian cancers[1][2][3][4][5][7].
