TAS1440 + ATRA is an investigational combination therapy being developed by Taiho Oncology for the treatment of relapsed or refractory acute myeloid leukemia (AML). TAS1440 is a small molecule inhibitor of lysine-specific demethylase 1 (LSD1), an epigenetic enzyme that plays a critical role in maintaining the undifferentiated state of AML blasts. All-trans retinoic acid (ATRA) is a vitamin A derivative that induces differentiation in myeloid cells. Preclinical studies suggest that LSD1 inhibition sensitizes non-promyelocytic AML cells to ATRA-induced differentiation, potentially overcoming the resistance typically seen in these subtypes. The combination is currently being evaluated in a Phase 1 clinical trial (NCT04282668) to assess safety, pharmacokinetics, and preliminary efficacy.
