TAS266 is a novel agonistic tetravalent nanobody targeting death receptor type 5 (DR5), also known as TRAIL receptor 2, with potential antineoplastic activity. The molecule consists of four single-domain heavy chain variable region (VHH) antibodies separated by peptide linkers, designed to cluster and activate DR5 on tumor cells, thereby inducing apoptosis. TAS266 demonstrated higher potency than cross-linked DR5 antibodies or TRAIL in preclinical studies. It was developed for the treatment of advanced solid tumors but was discontinued after phase I clinical trials due to unexpected but reversible hepatotoxicity attributed to immunogenicity and high activity against DR5-expressing hepatocytes[1][2][3][7].
