Tasonermin is a recombinant form of human tumor necrosis factor alpha (TNF-alpha), produced via Escherichia coli cell culture. It acts as a cytokine and exerts antitumor effects by binding to TNF receptors on the surface of cancer cells, particularly the Tumor necrosis factor receptor superfamily member 1A (TNFRSF1) receptor. This interaction initiates intracellular signaling cascades that lead to apoptosis through activation of caspases, resulting in programmed cell death. Tasonermin also has direct effects on tumor vasculature, promoting microvascular thrombosis and enhancing leukocyte infiltration into tumors. Indirectly, it modulates immune responses by activating monocytes, macrophages, granulocytes, and lymphocytes—enhancing both innate and adaptive immunity against tumor cells. Clinically, tasonermin is used in combination with melphalan for isolated limb perfusion in patients with soft tissue sarcoma of the limbs who are not candidates for resection or require palliative care. The drug was first approved in Europe under the brand name Beromun[2][3][5][6].
