TAT-0002 is an experimental targeted alpha therapy (TAT) currently in preclinical development for the treatment of prostate cancer, specifically metastatic castration-resistant prostate cancer (mCRPC). Developed by T-Alpha Therapeutics, the drug consists of a monoclonal antibody targeting the prostate-specific membrane antigen (PSMA) conjugated to the alpha-emitting radioisotope Actinium-225 (Ac-225). The mechanism of action involves the selective binding of the antibody to PSMA-expressing tumor cells, followed by internalization and the delivery of high-energy alpha radiation. This radiation induces potent double-strand DNA breaks within the target cells while minimizing damage to surrounding healthy tissue due to the short range of alpha particles. TAT-0002 is part of a pipeline of next-generation radiopharmaceuticals designed to overcome the limitations of beta-emitting therapies by providing higher linear energy transfer and improved efficacy in resistant tumors.
