Drug intelligence / Profile preview
TAT-CRYAB is a cell-penetrating recombinant fusion protein consisting of the HIV-1 Tat protein transduction domain (TAT) fused to alpha-crystallin B chain (CRYAB), a small heat shock protein. By utilizing the cell-penetrating peptide TAT, the fusion protein is able to cross cellular membranes and deliver CRYAB intracellularly. TAT-CRYAB acts as an anti-inflammatory and cytoprotective agent. In preclinical models, it has been shown to inhibit IKKβ activity, thereby suppressing NF-κB-mediated inflammatory responses and protecting mucosal barrier integrity. It also attenuates reactive oxygen species (ROS) generation and CaMKII activation. TAT-CRYAB is being investigated for the treatment of benign tracheal stenosis (BTS), inflammatory bowel disease (IBD) such as colitis, and diesel exhaust particle-induced cardiac arrhythmia.
