Overview

Tat-Hhi-ELP is a thermally responsive polypeptide conjugate designed to inhibit the Hedgehog (Hh) signaling pathway. Developed at the University of Mississippi Medical Center, the construct consists of a cell-penetrating peptide (Tat) and a Hedgehog-inhibitory peptide (Hhi) fused to an elastin-like polypeptide (ELP) biopolymer. The ELP component, derived from mammalian tropoelastin, undergoes a phase transition and aggregates at tumor sites in response to local hyperthermia, enabling targeted delivery. The Hhi peptide blocks the interaction of Sonic Hedgehog (Shh) ligand with the Patched-1 (PTCH1) receptor, thereby suppressing downstream Hh signaling and reducing GLI-1 transcription factor levels. Preclinical studies have demonstrated its efficacy in inhibiting the proliferation of breast cancer cells and targeting breast cancer stem cells.
