TAT0001 is an investigational targeted alpha therapy (TAT) being developed for the treatment of Glypican-3 (GPC3)-positive solid tumors, with a primary focus on hepatocellular carcinoma (HCC). The drug candidate consists of a high-affinity targeting vector, likely a monoclonal antibody or fragment, conjugated to the alpha-emitting radioisotope Actinium-225 (^225Ac). By binding specifically to GPC3, which is highly expressed on the surface of liver cancer cells but has limited expression in healthy adult tissues, TAT0001 delivers potent, localized alpha radiation directly to the tumor site. The high linear energy transfer (LET) and short range of alpha particles induce lethal double-stranded DNA breaks within the malignant cells while minimizing collateral damage to surrounding healthy parenchyma. This approach is designed to overcome the limitations of conventional therapies and beta-emitting radiopharmaceuticals in treating micro-metastatic and radioresistant tumors.
