Overview

TAT4 Gel is a topical formulation of the prostaglandin F2α (PGF2α) analog tafluprost, originally developed by Topokine Therapeutics and later acquired by Allergan (now part of AbbVie). It was designed for the non-invasive reduction of unwanted subcutaneous fat, specifically targeting the nasolabial folds (smile lines). The drug's mechanism of action involves the activation of the prostaglandin F (FP) receptor on adipocytes, which inhibits the differentiation of pre-adipocytes into mature fat cells (adipogenesis) and promotes the breakdown of lipids within existing adipocytes (lipolysis). This leads to a localized reduction in fat volume, a therapeutic application of the side effect known as prostaglandin-associated periorbitopathy observed in glaucoma patients treated with prostaglandin eye drops. TAT4 Gel reached Phase 2 clinical development for the reduction of moderate to severe nasolabial folds.
